Introduction

Maclusa Tablet is a fixed-dose combination tablet containing sofosbuvir and velpatasvir for oral administration. Each tablet
contains 400 mg sofosbuvir and 100 mg velpatasvir. Sofosbuvir is a nucleotide analog HCV NS5B polymerase inhibitor and
velpatasvir is an NS5A inhibitor.
Sofosbuvir:
The IUPAC name for sofosbuvir is (S)-Isopropyl 2-((S)-(((2R,3R,4R,5R)-5-(2,4-dioxo-3,4-dihydropyrimidin-1(2H)-yl)-4-
fluoro-3hydroxy-4-methyltetrahydrofuran-2yl)methoxy)(phenoxy)phosphorylamino)propanoate. It has a molecular
formula of C22H29FN3O9P and a molecular weight of 529.45.
Velpatasvir:
The IUPAC name for velpatasvir is Methyl {(1R)-2-[(2S,4S)-2-(5-{2-[(2S,5S)-1-{(2S)-2-[(methoxycarbonyl)amino]-3-
methylbutanoyl}-5-methylpyrrolidin-2-yl]-1,11-dihydro[2]benzopyrano[4′,3′:6,7]naphtho[1,2-d]imidazol-9-yl}-1Himidazol-2-yl)-4-(methoxymethyl)pyrrolidin-1-yl]-2-oxo-1-phenylethyl}carbamate. It has a molecular formula of
C49H54N8O8 and a molecular weight of 883.0.

Side Effects of Maclusa Tablet

  • Chest pain or discomfort.
  • large, hive-like swelling on the face, eyelids, lips, tongue, throat, hands, legs, feet, or sex organs.
  • lightheadedness, dizziness or fainting.
  • skin rash, sometimes with blisters or hive-like swelling.
  • slow or irregular heartbeat.
  • unconsciousness.
  • unusual tiredness.

Precautions

Before taking this medication, tell your doctor or pharmacist if you are allergic to sofosbuvir or velpatasvir; or if you have any other allergies. This product may contain inactive ingredients, which can cause allergic reactions or other problems. Talk to your pharmacist for more details.

Before using this medication, tell your doctor or pharmacist your medical history, especially of: other liver problems (such as hepatitis B), diabetes.

If you have diabetes, your blood sugar may be lower with hepatitis C treatment. This can increase your risk of low blood sugar, so your doctor may adjust your diabetes treatment plan. Tell your doctor right away if you have symptoms of low blood sugar such as sudden sweating, shaking, fast heartbeat, hungerblurred visiondizziness, or tingling hands/feet.

Before having surgery, tell your doctor or dentist about all the products you use (including prescription drugs, nonprescription drugs, and herbal products).

Children may be more sensitive to the side effects of this drug, especially vomiting.

During pregnancy, this medication should be used only when clearly needed. Discuss the risks and benefits with your doctor. Sofosbuvir/velpatasvir, in combination with ribavirin, must not be used during pregnancy by either the pregnant woman or her male partner. The combination may harm an unborn baby. Two reliable forms of birth control (such as condomsbirth control pills) must be used whenever at least one sexual partner is using these medicines together, and for six months after stopping treatment. If you or your partner becomes pregnant, or if you think you or your partner may be pregnant, tell your doctor right away.

It is unknown if this medication passes into breast milk. Consult your doctor before breast-feeding. Breast-feeding is not recommended if this medication is used in combination with ribavirin.
